NM_001110556.2(FLNA):c.2614G>A (p.Ala872Thr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A872T variant (also known as c.2614G>A), located in coding exon 17 of the FLNA gene, results from a G to A substitution at nucleotide position 2614. The alanine at codon 872 is replaced by threonine, an amino acid with similar properties. Based on data from gnomAD, the A allele has an overall frequency of 0.0025% (5/203047) total alleles studied, with 0 hemizygote(s) observed. The highest observed frequency was 0.0219% (4/18264) of African alleles. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
