In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with ATP7B-related disease. This variant is present in population databases (rs763053197, ExAC 0.002%). This sequence change replaces threonine with isoleucine at codon 1062 of the ATP7B protein (p.Thr1062Ile). The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and isoleucine.
