Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000022.11:g.(?_20043355)_(20064662_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 3-9 of the TANGO2 gene. The 5' boundary is likely confined to intron 2. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. Deletion of exons 3-9 has been reported to be homozygous or compound heterozygous in several individuals with rhabdomyolysis, metabolic crisis, and cardiac abnormalities (PMID: 26805781, 26805782). Loss-of-function variants in TANGO2 are known to be pathogenic (PMID: 26805781, 26805782). For these reasons, this variant has been classified as Pathogenic.
