Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.5080G>A (p.Glu1694Lys), citing Ambry Variant Classification Scheme 2023: The p.E1694K variant (also known as c.5080G>A), located in coding exon 20 of the AKAP9 gene, results from a G to A substitution at nucleotide position 5080. The glutamic acid at codon 1694 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
