Pathogenic for Oto-palato-digital syndrome, type II; Heterotopia, periventricular, X-linked dominant; Frontometaphyseal dysplasia; Melnick-Needles syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001110556.2(FLNA):c.3296_3304delinsTCGG (p.Gly1099fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in FLNA are known to be pathogenic (PMID: 16684786, 20730588, 26471271). This variant has been observed in an individual affected with clinical features of periventricular heterotopia (Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Gly1099Valfs*9) in the FLNA gene. It is expected to result in an absent or disrupted protein product.
