NM_020975.6(RET):c.2243G>A (p.Gly748Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G748D variant (also known as c.2243G>A), located in coding exon 12 of the RET gene, results from a G to A substitution at nucleotide position 2243. The glycine at codon 748 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on data from gnomAD, the frequency for this variant is above the maximum credible frequency for a MEN2-causing variant in this gene based on internally established thresholds (Karczewski et al. Nature. 2020 May;581(7809):434-443; Whiffin et al. Genet Med. 2017 10;19:1151-1158). Based on the supporting evidence, this variant is unlikely to be causative of MEN2; however, its contribution to the development of Hirschsprung disease is uncertain.
