NM_002439.5(MSH3):c.1000T>A (p.Tyr334Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y334N variant (also known as c.1000T>A), located in coding exon 6 of the MSH3 gene, results from a T to A substitution at nucleotide position 1000. The tyrosine at codon 334 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
