NM_177438.3(DICER1):c.3304A>G (p.Ile1102Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I1102V variant (also known as c.3304A>G), located in coding exon 20 of the DICER1 gene, results from an A to G substitution at nucleotide position 3304. The isoleucine at codon 1102 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
