Uncertain significance for Short QT syndrome type 3; Andersen Tawil syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000891.3(KCNJ2):c.101A>G (p.Lys34Arg), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 661299). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals affected with KCNJ2-related conditions. This variant is present in population databases (rs772815957, gnomAD 0.007%). This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 34 of the KCNJ2 protein (p.Lys34Arg). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
