Uncertain significance for Primary dilated cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001318895.3(FHL2):c.533A>C (p.Glu178Ala), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C25"). This sequence change replaces glutamic acid with alanine at codon 178 of the FHL2 protein (p.Glu178Ala). The glutamic acid residue is moderately conserved and there is a moderate physicochemical difference between glutamic acid and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FHL2-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
