This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 533, where C is replaced by T; at the protein level this means replaces proline at residue 178 with leucine — a missense variant. Submitter rationale: Variant summary: ATM c.533C>T (p.Pro178Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 243558 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.533C>T has been reported in the literature in one individual affected with AML (Choi_2018).
