NM_001927.4(DES):c.65C>G (p.Pro22Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P22R variant (also known as c.65C>G), located in coding exon 1 of the DES gene, results from a C to G substitution at nucleotide position 65. The proline at codon 22 is replaced by arginine, an amino acid with dissimilar properties. This variant has been reported in a dilated cardiomyopathy (DCM) cohort (Zhang XL et al. Sci Rep, 2020 Feb;10:2226). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
