Uncertain significance for METHYLMALONIC ACIDURIA AND HOMOCYSTINURIA, cblJ TYPE — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005050.4(ABCD4):c.1352T>C (p.Phe451Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with serine at codon 451 of the ABCD4 protein (p.Phe451Ser). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ABCD4-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15").
