Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.1934A>T (p.Gln645Leu), citing Ambry Variant Classification Scheme 2023: The p.Q645L variant (also known as c.1934A>T), located in coding exon 7 of the BLM gene, results from an A to T substitution at nucleotide position 1934. The glutamine at codon 645 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
