Uncertain significance for Cystic fibrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000492.4(CFTR):c.932T>G (p.Phe311Cys), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has been reported to have conflicting or insufficient data to determine the effect on CFTR protein function (PMID: 28242630). This variant has not been reported in the literature in individuals with CFTR-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces phenylalanine with cysteine at codon 311 of the CFTR protein (p.Phe311Cys). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and cysteine.
