This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 1053, where G is replaced by T; at the protein level this means replaces methionine at residue 351 with isoleucine — a missense variant. Submitter rationale: This sequence change replaces methionine with isoleucine at codon 351 of the MSH2 protein (p.Met351Ile). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and isoleucine. This variant is present in population databases (rs373122667, ExAC 0.01%). This variant has not been reported in the literature in individuals with MSH2-related disease.
