Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000890.5(KCNJ5):c.260G>A (p.Arg87His), citing Ambry Variant Classification Scheme 2023: The p.R87H variant (also known as c.260G>A), located in coding exon 1 of the KCNJ5 gene, results from a G to A substitution at nucleotide position 260. The arginine at codon 87 is replaced by histidine, an amino acid with highly similar properties, and is located in the TM domain. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
