Uncertain significance for Lethal multiple pterygium syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000751.3(CHRND):c.314C>G (p.Pro105Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with CHRND-related disease. This variant is present in population databases (rs769160054, ExAC 0.006%). This sequence change replaces proline with arginine at codon 105 of the CHRND protein (p.Pro105Arg). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and arginine.
