Uncertain significance for Charcot-Marie-Tooth disease axonal type 2L — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014365.3(HSPB8):c.502G>A (p.Ala168Thr), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 168 of the HSPB8 protein (p.Ala168Thr). This variant has not been reported in the literature in individuals affected with HSPB8-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 659622).
