Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.7919A>G (p.Tyr2640Cys), citing Ambry Variant Classification Scheme 2023: The p.Y2619C variant (also known as c.7856A>G), located in coding exon 53 of the NF1 gene, results from an A to G substitution at nucleotide position 7856. The tyrosine at codon 2619 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
