NM_025137.4(SPG11):c.2008G>A (p.Val670Ile) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V670I variant (also known as c.2008G>A), located in coding exon 10 of the SPG11 gene, results from a G to A substitution at nucleotide position 2008. The valine at codon 670 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
