Submitter rationale: ABCB11 c.150+3A>C is an intronic variant located in the donor splice region of intron 4. This variant has been observed in at least one proband with an ABCB11-related disorder (PMID:35626323;32087350;27114171). At least one splicing study has demonstrated that this variant results in aberrant splicing (PMID:27114171). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB11 c.150+3A>C as a pathogenic variant.
