This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 1525, where T is replaced by C; at the protein level this means replaces serine at residue 509 with proline — a missense variant. Submitter rationale: This sequence change replaces serine with proline at codon 509 of the SDHA protein (p.Ser509Pro). The serine residue is moderately conserved and there is a moderate physicochemical difference between serine and proline. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SDHA-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
