Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.7232CAC[7] (p.Pro2414_Pro2415dup), citing Ambry Variant Classification Scheme 2023: The c.7241_7246dupCACCAC variant (also known as p.P2414_P2415dup), located in coding exon 34 of the NOTCH1 gene, results from an in-frame duplication of CACCAC at nucleotide positions 7241 to 7246. This results in the duplication of 2 extra residues (PP) between codons 2414 and 2415. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
