Submitter rationale: The p.E462* pathogenic mutation (also known as c.1384G>T), located in coding exon 9 of the BRCA2 gene, results from a G to T substitution at nucleotide position 1384. This changes the amino acid from a glutamic acid to a stop codon within coding exon 9. This variant has been previously detected in an African American patient diagnosed with breast cancer under the age of 40 (Rummel SK et al. Breast Cancer Res. Treat., 2017 Aug;164:593-601). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD).
