Uncertain significance for Autosomal recessive distal spinal muscular atrophy 1; Charcot-Marie-Tooth disease axonal type 2S — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002180.3(IGHMBP2):c.1301T>G (p.Val434Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine with glycine at codon 434 of the IGHMBP2 protein (p.Val434Gly). The valine residue is moderately conserved and there is a moderate physicochemical difference between valine and glycine. This variant is not present in population databases (ExAC no frequency). This variant has been observed on the opposite chromosome (in trans) from a pathogenic variant in an individual affected with Charcot-Marie-Tooth disease (Invitae). This finding is consistent with autosomal recessive inheritance, and suggests that this variant contributes to disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
