This variant lies in the PLOD1 gene (transcript NM_000302.4) at coding-DNA position 315 through coding-DNA position 317, deleting 3 bases; at the protein level this means deletes phenylalanine at residue 106. Submitter rationale: The c.315_317delGTT variant (also known as p.F106del) is located in coding exon 4 of the PLOD1 gene. This variant results from an in-frame GTT deletion at nucleotide positions 315 to 317. This results in the in-frame deletion of a phenylalanine at codon 106. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
