Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.(?_31200422)_(31265339_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 9-35 of the NF1 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Similar deletions of exons 9-35 have been observed in an individual affected with clinical features of neurofibromatosis type 1 (PMID: 16283621). Exons 9-35 are also referred to as exons 7-27b in the literature. Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). For these reasons, this variant has been classified as Pathogenic.
