Pathogenic for Duchenne muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.11:g.(?_31968329)_(31968524_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 45 of the DMD gene. This sequence change is predicted to create a premature translational stop signal and result in an absent or disrupted protein product. However, experimental studies have shown that it may also lead to a change in mRNA splicing that causes skipping of exon 44, which results in an in-frame deletion and preserves the reading frame (PMID: 9040743, 24871807). This variant has been reported in multiple individuals with both Duchenne muscular dystrophy and Becker muscular dystrophy (PMID: 11257468, 18683213, 25244321, 26081009). For these reasons, this variant has been classified as Pathogenic.
