Pathogenic for Familial adenomatous polyposis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.10:g.(?_112737024)_(112844136_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing promoter 1B, promoter 1A, and the full coding sequence of the APC gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Gross deletions of APC have been observed in individuals affected with familial adenomatous polyposis, including de novo cases, however paternity was not confirmed (PMID: 18487285,Â¬â€ 19279422,Â¬â€ 21643010,Â¬â€ 26681312). Loss-of-function variants in APC are known to be pathogenic (PMID: 20685668, 17963004). For these reasons, this variant has been classified as Pathogenic.
