Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_058216.3(RAD51C):c.460G>A (p.Glu154Lys), citing Ambry Variant Classification Scheme 2023: The p.E154K variant (also known as c.460G>A), located in coding exon 3 of the RAD51C gene, results from a G to A substitution at nucleotide position 460. The glutamic acid at codon 154 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
