Uncertain significance for Familial acute necrotizing encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006267.5(RANBP2):c.6881A>T (p.Asp2294Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with valine at codon 2294 of the RANBP2 protein (p.Asp2294Val). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and valine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals with RANBP2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
