NM_032638.5(GATA2):c.1242C>G (p.Phe414Leu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F414L variant (also known as c.1242C>G), located in coding exon 5 of the GATA2 gene, results from a C to G substitution at nucleotide position 1242. The phenylalanine at codon 414 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
