Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.2923C>G (p.Gln975Glu), citing Ambry Variant Classification Scheme 2023: The p.Q975E variant (also known as c.2923C>G), located in coding exon 14 of the BLM gene, results from a C to G substitution at nucleotide position 2923. The glutamine at codon 975 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
