This variant has been observed in an individual with clinical features of hyperekplexia (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with valine at codon 637 of the SLC6A5 protein (p.Ala637Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine.
