Uncertain significance for Glycogen storage disease type III — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000642.3(AGL):c.4382C>G (p.Ala1461Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with glycine at codon 1461 of the AGL protein (p.Ala1461Gly). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and glycine. This variant is present in population databases (rs749209422, ExAC 0.03%). This variant has not been reported in the literature in individuals with AGL-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
