This variant has not been reported in the literature in individuals with WASHC5-related conditions. This variant is present in population databases (rs766713240, ExAC 0.003%). This sequence change replaces serine with proline at codon 433 of the WASHC5 protein (p.Ser433Pro). The serine residue is moderately conserved and there is a moderate physicochemical difference between serine and proline.
