Uncertain significance for Nephrolithiasis/nephrocalcinosis — the classification assigned by Ambry Genetics to NM_000388.4(CASR):c.3112G>A (p.Gly1038Arg), citing Ambry Variant Classification Scheme 2023: The p.G1038R variant (also known as c.3112G>A), located in coding exon 6 of the CASR gene, results from a G to A substitution at nucleotide position 3112. The glycine at codon 1038 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
