Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004100.5(EYA4):c.251G>A (p.Ser84Asn), citing Ambry Variant Classification Scheme 2023: The p.S84N variant (also known as c.251G>A), located in coding exon 4 of the EYA4 gene, results from a G to A substitution at nucleotide position 251. The serine at codon 84 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
