Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003002.4(SDHD):c.436G>A (p.Asp146Asn), citing Ambry Variant Classification Scheme 2023: The p.D146N variant (also known as c.436G>A), located in coding exon 4 of the SDHD gene, results from a G to A substitution at nucleotide position 436. The aspartic acid at codon 146 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
