NM_001166108.2(PALLD):c.2251G>A (p.Glu751Lys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E734K variant (also known as c.2200G>A) is located in coding exon 12 of the PALLD gene. The glutamic acid at codon 734 is replaced by lysine, an amino acid with similar properties. This change occurs in the first base pair of coding exon 12. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
