Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_017534.6(MYH2):c.5472+3G>T, citing LabCorp Variant Classification Summary - May 2015: Variant summary: MYH2 c.5472+3G>T alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Computational tools predict a significant impact on normal splicing: Two predict the variant weakens a canonical 5' donor site. One predicts the variant has no significant impact on splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 9.1e-05 in 251424 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for disease-causing variants in MYH2, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.5472+3G>T in individuals affected with MYH2-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 654743). Based on the evidence outlined above, the variant was classified as uncertain significance.
