Pathogenic for Tuberous sclerosis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.10:g.(?_2050390)_(2050496_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an in-frame deletion of the genomic region encompassing exon 3 of the TSC2 gene. It preserves the integrity of the reading frame. A gross deletion of exon 3 has been reported to be de novo in an individual affected with tuberous sclerosis in the Leiden Open-source Variation Database (PMID: 21520333). Due to alternative exon numbering, a deletion of exon 3 may also be referred to as a deletion of exon 2. ClinVar contains an entry for a deletion of exon 3 (Variation ID: 467839). Experimental studies and prediction algorithms are not available for this variant, and the functional significance of the affected amino acid(s) is currently unknown. For these reasons, this variant has been classified as Pathogenic.
