This variant lies in the COL6A3 gene (transcript NM_004369.4) at coding-DNA position 6832, where C is replaced by A; at the protein level this means replaces proline at residue 2278 with threonine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
