Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001271696.3(ABCB7):c.1199T>C (p.Ile400Thr), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with ABCB7-related disease. This sequence change replaces isoleucine with threonine at codon 401 of the ABCB7 protein (p.Ile401Thr). The isoleucine residue is highly conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is not present in population databases (ExAC no frequency).
