Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.1799A>G (p.Glu600Gly), citing Ambry Variant Classification Scheme 2023: The p.E600G variant (also known as c.1799A>G), located in coding exon 11 of the FLNA gene, results from an A to G substitution at nucleotide position 1799. The glutamic acid at codon 600 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
