Pathogenic for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014874.4(MFN2):c.262A>T (p.Ile88Phe), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MFN2 gene (transcript NM_014874.4) at coding-DNA position 262, where A is replaced by T; at the protein level this means replaces isoleucine at residue 88 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with phenylalanine at codon 88 of the MFN2 protein (p.Ile88Phe). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with autosomal dominant Charcot-Marie-Tooth disease (PMID: 26392352, Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 653992). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). For these reasons, this variant has been classified as Pathogenic.
