NM_002691.4(POLD1):c.3158G>A (p.Arg1053His) was classified as Uncertain significance for Colorectal cancer, susceptibility to, 10 by KCCC/NGS Laboratory, Kuwait Cancer Control Center, citing ACMG Guidelines, 2015: For the POLD1 variant, the sequence change replaces arginine with histidine at codon 1053 of the POLD1 protein (p.Arg1053His). The arginine residue is highly conserved and there is a small physicochemical difference between arginine and histidine. This variant is present in population databases (ExAC frequency 0.004%). This variant has not been reported in the literature in individuals with POLD1-related disease. In-silico simulator softwares to predict the effect of missense changes on protein structure and function showed (SIFT: deleterious; PolyPhen-2: probably damaging). Therefore, it has been classified as a Variant of Uncertain Significance.
