Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.428C>A (p.Ser143Tyr), citing Ambry Variant Classification Scheme 2023: The p.S143Y variant (also known as c.428C>A), located in coding exon 4 of the RAD50 gene, results from a C to A substitution at nucleotide position 428. The serine at codon 143 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
