Uncertain significance for Dyskeratosis congenita, autosomal dominant 2; Idiopathic Pulmonary Fibrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198253.3(TERT):c.2915G>A (p.Arg972His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 972 of the TERT protein (p.Arg972His). This variant is present in population databases (no rsID available, gnomAD 0.003%). This missense change has been observed in individual(s) with clinical features of autosomal dominant pulmonary fibrosis and/or dyskeratosis congenita (PMID: 21931702, 29749397). ClinVar contains an entry for this variant (Variation ID: 653306). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The histidine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
